Isolation and Development of Highly Polymorphic Microsatellites in Rhacophorus hui

Abstract


Rhacophorus hui and Rhacophorus dugritei were once considered as parts of the same species for their morphological similarity, and subsequent studies found the chromosomal inversions in their genome made them separated into two different species finally. To explore the relationship between the two species and perform population genetics analysis of R. hui, highly polymorphic microsatellite markers were required. Here we reported the isolation and characterization of thirteen highly polymorphic novel microsatellite markers of R. hui for phylogenetic and population genetic analysis. The number of alleles per locus ranged from 12 to 29, and the average heterozygosity observed and expected per locus ranged from 0.429 to 0.930 and 0.848 to 0.959, respectively. As for sequential Bonferroni corrections, three pairs showed significant departure from Hardy-Weinberg equilibrium. These polymorphic loci will be useful markers in the research of R. hui.
